Scientific data (oral beta-1,3/1,6-glucan from yeast)
A systematic review and meta-analysis of 13 randomized placebo-controlled trials in healthy subjects showed that yeast β-glucans statistically significantly reduced the incidence/risk of URTI episodes (OR = 0.345; 95% CI 0.192–0.620), and also reduced the number of episodes (SMD = −0.315) and duration of cold episodes (SMD = −0.312). The authors also noted that in most of the included studies, β-glucan improved symptom severity , and the supplement was generally well tolerated .
In a placebo-controlled study in marathon runners, Wellmune WGP® (β-1,3/1,6-glucan from yeast) at a dose of 250 or 500 mg/day for 4 weeks after the race was associated with a lower incidence of URTI symptoms: at week 4, URTI was reported by 8% of participants in the β-glucan groups versus 24% in the placebo group.
Another study after intense exercise showed that β-glucan was associated with a 37% reduction in the number of “cold/flu symptom days” after a marathon (compared to placebo) and a 32% increase in secretory IgA in saliva 2 hours after exercise, a marker of mucosal immune defense of the respiratory tract.
In the RCT with a beverage containing dispersed yeast β-glucan, fewer symptomatic days (3.43 ± 6.44 vs. 3.84 ± 6.84) and lower total symptom severity (4.52 ± 1.61 vs. 5.60 ± 2.23) were reported in the per-protocol analysis, as well as fewer missed training days due to URTI after a marathon (0.09 ± 0.38 vs. 0.36 ± 1.40).

Mechanism of action of β-1,3/1,6-glucan
β-1,3/1,6-glucan from the yeast cell wall is considered a molecular “signal” (MAMP) that is recognized by innate immune receptors on defense cells. Dectin-1 is considered a key receptor (on macrophages, neutrophils, dendritic cells), and interaction/cooperation with CR3 and other PRRs is described - this can trigger intracellular signaling cascades (in particular via Syk/CARD9 → NF-κB) and maintain the “readiness” of cells to respond to microbial stimuli. What happens when ingested: Some of the β-glucan particles pass into the upper small intestine, where they can be taken up by cells of the intestinal immune system (macrophages/dendritic cells). The β-glucan is then phagocytized and processed , and the smaller soluble fragments can then be transported to immune organs (e.g., spleen) where they prime other innate immune cells—one of the most common mechanistic scenarios for the systemic effects of oral ingestion. “Trained immunity”: In modern reviews, β-glucans are described as one of the classic triggers of a longer-term functional reprogramming of innate immune cells (mainly monocytes/macrophages) — after a “first contact”, the cells can respond faster/more effectively to repeated stimuli. This effect is associated with receptor pathways (in particular Dectin-1) and intracellular changes in response regulation. [1–2,5]
- Summary for practice: β-1,3/1,6-glucan works not as a “vitamin,” but as a component that can gently modulate the activity of innate immunity — especially important during periods of stress, seasonal changes, and stress.
